首先,一个问题是,有时运行它(在IDLE中使用F5)将不会显示任何内容。只需“>>”,我就必须关闭shell并重新运行它。

其次,在赢得并拒绝再次播放的提议后,仍然要求输入猜测。我想while循环仍然会运行,所以我怎么能阻止它呢?

这段代码最大的问题是你到处都在使用全局变量;如果你在python对象中存储值并在每次开始新游戏时初始化你的对象,这会好得多。

要修复您现在使用的内容,您需要在letters函数中初始化play_again() ...
